Immunophenotyping of peripheral blood mononuclear cells in tuberous sclerosis complex

چکیده

Abstract Tuberous sclerosis complex (TSC) is a multisystem disorder caused by loss-of-function mutations in TSC1/2 proteins. This results constitutive mTOR activation and predisposes to the development of benign tumours, with renal angiomyolipomas (AMLs) tuber-related drug-resistant epilepsy (DRE) being major causes morbidity mortality adults. Despite central role leukocyte biology beneficial impact inhibitors TSC, immune system inflammation tumours DRE people TSC remains unknown. Using peripheral blood mononuclear cells (PBMCs) serum from healthy controls (HC) non-TSC DRE, we employed flow cytometry multiplex assays interrogate profile associated chronic loss inhibition. We found that higher levels astroglial marker GFAP are elevated show high intergenotype variability. also observed differences circulating cell populations, cytokines, chemokines growth factors related TSC. proportion B parallel increased total IgG but reduction IL-2 IL-7 lower frequency T cells. vs without AML showed CD4:CD8 ratio, monocytes dendritic frequencies. Finally, affected polarization, leading an c17CD8 +T cells, especially subjects DRE. Overall, our findings indicate biomarkers for inflammation, glial activation, distribution PBMCs compared controls.

Toll Like Receptor 2 and 4 Expression in Peripheral Blood Mononuclear Cells of Multiple Sclerosis Patients

Background: Multiple sclerosis (MS) is a T cell mediated autoimmune disease with unknown etiology. Appropriate MS therapeutic strategies need thorough understanding of both disease etiology and pathogenesis mechanisms. Ligation of TLR-2 and TLR-4 stimulates the production of several cytokines leading to CNS autoimmunity and neurodegenerative diseases. Objective: To find a relationship between M...

Optimization of human peripheral blood mononuclear cells (PBMCs) cryopreservation

Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...

Interleukin-17A and Interleukin-17F mRNA Expressions in Peripheral Blood Mononuclear Cells of Patients with Multiple Sclerosis

Bakground: Multiple sclerosis (MS) is a CD4+ T cell-mediated autoimmune disease affecting the central nervous system (CNS). It was previously believed that Th1 cells were pathogenic T cells in experimental autoimmune encephalomyelitis (EAE). However, the functional role of Th1 cells in EAE has been reconsidered upon the discovery of IL-17- producing T cells which are consider as dominant effect...

Amyotrophic Lateral Sclerosis Multiprotein Biomarkers in Peripheral Blood Mononuclear Cells

BACKGROUND Amyotrophic lateral sclerosis (ALS) is a fatal progressive motor neuron disease, for which there are still no diagnostic/prognostic test and therapy. Specific molecular biomarkers are urgently needed to facilitate clinical studies and speed up the development of effective treatments. METHODOLOGY/PRINCIPAL FINDINGS We used a two-dimensional difference in gel electrophoresis approach...
